Toward a serum-free, xeno-free culture system for optimal growth and expansion of hMSC suited to therapeutic applications

چکیده
Background Human mesenchymal stem cells (hMSC) hold great promise as a tool in regenerative medicine and cell therapy. Application of hMSC in cell therapy requires the elaboration of an appropriate serum-free (SF), xeno-free (XF) culture system in order to minimize the health risk of using xenogenic compounds, and to limit the immunological reactions in-vivo. Besides the well-known disadvantages of serum, in comparison to a SF, XF culture system, serum also exhibits poor performance in the context of hMSC proliferation. In the present study, a novel SF, XF culture system for hMSC suitable for therapeutic applications was developed and evaluated. The SF, XF culture system includes specially developed solutions for attachment, dissociation, and freezing, as well as a culture medium, MSC NutriStem XF, that enables long-term growth of multipotent hMSC. Development of the SF, XF culture system was conducted on hMSC from a variety of sources: bone marrow (BM), adipose tissue (AT) and Wharton’s jelly (WJ).

Human mesenchymal stem/stromal cells (hMSCs) have generated great interest in regenerative medicine mainly due to their multidifferentiation potential and immunomodulatory role. Although hMSC can be obtained from different tissues, the number of available cells is always low for clinical applications, thus requiring in vitro expansion. Most of the current protocols for hMSC expansion make use o...
متن کاملLong Term Expansion of Bone Marrow-Derived hMSCs on Novel Synthetic Microcarriers in Xeno-Free, Defined Conditions
Human mesenchymal stem cells (hMSCs) present an attractive target for cell therapy given their wide availability, immunomodulatory properties, and multipotent nature for differentiation into chondrocytes, osteocytes, and adipocytes. With the progression of hMSC clinical studies, there is an increasing demand for development of technologies that enable efficient cell scale-up into clinically rel...

Background: Large scale high quality and low cost production of clinical grade mesenchymal stem cells (MSCs) is critical for facilitating clinical research and potential stem cell therapies. Use of Fetal Bovine/Calf serum (FBS/FCS) for the clinical application carries a risk for transmission of zoonosis, therefore alternative xeno-free cell culture techniques have been evaluated. The commercial...

Human mesenchymal stem cells (HMSCS) possess three properties of great interest for the development of cell therapies and tissue engineering: multilineage differentiation, immunomodulation, and production of trophic factors. Efficient ex vivo expansion of hMSCs is a challenging requirement for large scale production of clinical grade cells. Low-cost, robust, scalable culture methods using chemi...

The manufacture of clinical grade cellular products for adoptive immunotherapy requires ex vivo culture and expansion of human T cells. One of the key components in manufacturing of T cell therapies is human serum (HS) or fetal bovine serum (FBS), which can potentially expose immunotherapy recipient to adventitious infectious pathogens and are thus considered as non-cGMP compliant for adoptive ...
